Abstract: PURPOSE: Paediatric populations with epilepsy offer a specific challenge in dose individualisation since they undergo significant developmental changes from childhood to adulthood. Population pharmacokinetic modelling is able to analyze sparse data sets such as that frequently available in paediatric populations. This study investigated the population pharmacokinetics of lamotrigine, in a paediatric population.
METHOD: In this study, pharmacokinetic models were developed using nonlinear mixed effects modelling, NONMEM, and used in the individualisation of new-generation anti-epileptic drug therapy in paediatric epilepsy. These models included the influence of covariates such as drug plasma concentration, age, body mass index and co-administered antiepileptic drugs.
