Abstract: We read with interest the letter by Hézard et al. regarding a modified thrombin generation (TG) assay using Protac® as a screening tool for protein C (PC) deficiency. We have developed a similar modification in order to make the TG- assay more sensitive to changes in the protein C pathway. We tested 122 patients (32 controls, 38 heterozygous for factor V Leiden, 34 family members of patients with an inherited thrombophilia with normal thrombophilia screens, 8 with PC deficiency and 10 with protein S deficiency) enrolled in a previous thrombophilia study. Although our assay is similar to the assay described by Hézard et al., we feel that there are important differences that impact on the reproducibility and accuracy of this test.
